Як уникнути eskape патогенів використовуючи екстракти їстівних рослин
The aim of the work was to determine the content of biologically active substances, namely polyphenols and anthocyanins, in the extracts of cherryplum, blueberries, jostaberry, sweet cherries, plums, red and black currants, and to study the effect of these extracts on the growth and biofilm formation of clinical isolates of ESKAPE Pathogens in vitro.Materials and methods. The antibiotic resistance of the following clinical isolates: Enterococcus faecalis, Staphylococcus aureus, Klebsiella pneumoniae, Acinetobacter baumannii, Pseudomonas aeruginosa, Enterobacter cloacae was determined by the Kirby-Bauer diffusion method. The bioactive substances content was determined with a thin layer chromatography method. The effect of berry extracts on the above-mentioned isolates was studied by the method of compatible cultivation. The capacity of clinical isolates to form a biofilm was studied by a spectrophotometric method, using gentian violet.Results. Having analyzed the results of the antibiotic sensitivity of clinical isolates, it was found, that they were resistant to all antimicrobials used. The analysis of the content of bioactive substances of berry extracts showed that they contain a large amount of anthocyanins and polyphenols. It was revealed, that clinical isolates are capable of forming biofilms, and the selected berry extracts had the ability to inhibit the formed biofilms with isolates of clinical origin, such as Klebsiella pneumoniae and Pseudomonas aeruginosa.Conclusions. Researching the antibacterial properties of anthocyanins and polyphenols, extracted from berries, we can conclude, that they are capable of inhibiting the growth of not only planktonic strains forms, selected by us, but also inhibit the biofilms, formed by themЦелью работы было определение содержания биологически активных веществ, а именно полифенолов и антоцианов, в экстрактах плодов алычи, черники, йошты, черешни, сливы, красной и черной смородины, и исследование влияния экстрактов плодов этих растений на рост и биопленкообразование клинических изолятов «возбудителей ESKAPE» in vitro.Материалы и методы. Антибиотикорезистентность штаммов клинического происхождения, а именно Enterococcus faecalis, Staphylococcus aureus, Klebsiella pneumoniae, Acinetobacter baumannii, Pseudomonas aeruginosa, Enterobacter cloacae определяли методом диффузии Кирби-Бауэра. Состав биологически активных веществ определяли методом тонкослойной хроматографии. Влияние экстрактов на вышеупомянутые изоляты изучали методом их совместного культивирования. Способность клинических изолятов к образованию биопленки изучали с помощью спектрофотометрического метода с использованием кристаллического фиолетового, в качестве красителя.Результаты. Проанализировав результаты чувствительности к антибиотикам клинических изолятов, было установлено, что они устойчивы ко всем антимикробным препаратам. Анализ содержания биологически активных веществ экстрактов ягод показал, что они содержат большое количество антоцианов и полифенолов, и обладают антибактериальными свойствами. Было установлено, что изоляты клинического происхождения способны к формированию биопленок, а отобранные нами экстракты ягод обладают способностью ингибировать сформированы биопленки изолятами клинического происхождения, а именно: Klebsiella pneumoniae и Pseudomonas aeruginosa.Выводы. Исследуя антибактериальные свойства антоцианов и полифенолов, экстрагированных из ягод, можно сделать вывод, что они способны ингибировать рост не только выделенных нами планктонных форм штаммов, но и подавляют образованные ими биопленкиМетою роботи було визначення вмісту біологічно активних речовин, а саме поліфенолів та антоціанів,в екстрактах плодів аличі, чорниці, йошти, черешні, сливи, червоної та чорної смородини, та дослідження впливу екстрактів плодів цих рослин на ріст та біоплівкоутворенняклінічних ізолятів "збудників ESKAPE"in vitro.Матеріали і методи. Антибіотикорезистентність штамів клінічного походження, а саме Enterococcus faecalis, Staphylococcus aureus, Klebsiella pneumoniae, Acinetobacter baumannii, Pseudomonas aeruginosa, Enterobacter cloacae визначали методом дифузії Кірбі-Бауера. Вміст біологічно активних речовин визначали методом тонкошарової хроматографії. Вплив ягідних екстрактів на вищезгадані ізоляти вивчали методом їх сумісного культивування. Здатність клінічних ізолятів до утворення біоплівки вивчали за допомогою спектрофотометричного методу з використанням кристалічного фіолетового, в якості барвника.Результати. Проаналізувавши результати чутливості до антибіотиків клінічних ізолятів, було встановлено, що вони стійкі до всіх антимікробних препаратів. Аналіз вмісту біологічно активних речовин екстрактів ягід показав, що вони містять велику кількість антоціанів та поліфенолів, та володіють антибактеріальними властивостями. Було встановлено, що ізоляти клінічного походження здатні до біоплівкоутворення, а відібрані нами екстракти ягід володіють здатністю інгібувати сформовані біоплівки ізолятами клінічного походження, а саме: Klebsiella pneumoniae та Pseudomonas aeruginosa.Висновки. Досліджуючи антибактеріальні властивості антоціанів та поліфенолів, екстрагованих з ягід, можна зробити висновок, що вони здатні гальмувати ріст не лише виділених нами планктонних форм штамів, але й пригнічують утворені ними біоплівк

Екстракти гранату, хурми, кропиви, кропу, капусти кале і Sideritis специфічно модулюють мікробіоту кишечникута локальну продукцію цитокінів в експериментах in vivo
The aim of the work was to use the in vivo model to reveal main changes in gut microbiota of immune competent mice in dynamic and find out the specificity of immunomodulation action (activity) of traditional foods ingredients - edible plants extracts on local mucosal cytokines to its oral administration.Materials and methods. In this study, seven groups of immunocompetent BALB/c mice were formed. All experimental mice had been fed orally by plants’ extracts (15mg/200 µl/mouse) for the 14th days. The extracts of edible plants - ingredients of traditional food such as kale leaves, persimmon, pomegranate, dill, Sideritis scardica, and nettle were obtained as described and orally administrated to experimental animals. For microbiological analysis of gut microbiota changes, the colon content has been investigated, the key microbial representatives were isolated by plating of its serial dilution on selected chromogenic medium, identified serologically and biochemically. The production of cytokines in different gut compartments and gut associated lymphoid tissues (GALT) were detected by Enzyme-linked immunosorbent assay (ELISA).Results. In experiments in mice, the ability of Kale, Dill and Sideritis extracts, when administered orally, selectively inhibit the content of E. coli, K. pneumoniae, E. faecalis, L. acidophilus in the colon of mice had been demonstrated, and increasing of B. bifidum had been observed. Nettle extract leads to an increase in E. coli, and persimmon extract - to an increase in levels of E. faecalis, Bifidobacterium and a decrease in the content of Candida spp. Pomegranate extract specifically stimulates the growth of Bifidobacterium. There are sufficient differences in produced cytokines in fragment culture and serum of mice fed with different plants extracts. TNF-a, and IL-2 increased both systemically and locally in the different gut compartments by Dill extract, Nettle and Sideritis extracts only at mucosal sites. IL-2, but also IL-10 and IL-12, IFN-g, and IL-17 but not TNF-a were stimulated in different levels by Pomegranate, Persimmon and Kale extracts: both systemically and locally.Conclusions. No harmful influence of tested plants had been observed. The most beneficial properties are inherent to Persimmon extract and slightly less detected in Pomegranate and Kale extracts. В исследовании было сформировано семь групп иммунокомпетентных мышей BALB/c. Все экспериментальные мыши в течение 14 дней перорально выпаивались экстрактами растений (15 мг/200 мкл/мышь). Экстракты съедобных растений - ингредиенты традиционных продуктов питания, такие как листья капусты, хурма, гранат, укроп, Sideritis scardica и крапива, были получены как описано и перорально введены экспериментальным животным. Для микробиологического анализа изменений кишечной микробиоты исследовали содержимое толстой кишки, ключевые представители микробиоты были изолированы путем высева серийных разведений на селективные хромогенные питательные среды, идентифицированы серологически и биохимически. Продукцию цитокинов различными отделами кишечника и лимфатической ткани, ассоциированной со слизистыми оболочками кишечника (КАЛТ), определяли методом иммуноферментного анализа (ИФА).Результаты. В экспериментах на мышах доказана способность экстрактов капусты кале, укропа и Sideritis при их оральном применении избирательно ингибировать E. сoli, K. pneumoniae, E. faecalis, L. acidophilus в толстой кишке мышей, способствуя при этом одновременному увеличению количества B. bifidum. Экстракт крапивы приводит к увеличению количества E. coli, а экстракт хурмы - к росту популяционных уровней E. faecalis, Bifidobacterium и уменьшение содержания Candida spp. Экстракт граната специфически стимулирует рост Bifidobacterium. Существуют значительные различия в продуцируемых цитокинах в супернатантах культуры фрагментов тканей и в сыворотке крови мышей при оральном введении им различных растительных экстрактов. Экстракт укропа предопределял системное и локальное увеличение ФНО-a и ИЛ-2, экстракты крапивы и Sideritis - на участках слизистой оболочки. ИЛ-2, ИЛ-10 и ИЛ-12, ИФН-γ и ИЛ-17, но не ФНО-a стимулировались на разных уровнях экстрактами граната, хурмы и капусты: как системно, так и локально.Выводы. Не наблюдали вредного воздействия тестируемых растений. Наиболее полезными свойствами обладает экстракт хурмы, экстракты граната и капусты обладают менее полезными свойствами. У дослідженні було сформовано сім груп імунокомпетентних мишей BALB/c, які протягом 14 днів перорально випоювались рослинними екстрактами (15 мг/200 мкл/миша). Останні одержували за розробленою нами технологією із харчових продуктів рослинного походження – капусти кале, кропу, Sideritis scardica, кропиви, плодів хурми і гранату. Матеріалом мікробіологічного аналізу змін кишкової мікробіоти слугував вміст товстої кишки. Ключових представників мікробіоти ізолювали шляхом висіву серійних розведень на селективні хромогенні поживні середовища та ідентифікували серологічно і біохімічно. Продукцію цитокінів різними відділами кишечнику та лімфатичної тканини, асоційованої із слизовими оболонками кишечнику, визначали методом імуноферментного аналізу.Результати. В експериментах на мишах доведено здатність екстрактів капусти кале, кропу і Sideritis при їх оральному застосуванні вибірково інгібувати вміст E. сoli, K. pneumoniae, E. faecalis, L. acidophilus у товстій кишці мишей, стимулюючи при цьому ріст B. bifidum. Показано, що екстракт кропиви призводить до збільшення кількості E. coli, а екстракт хурми – до зростання популяційних рівнів E. faecalis, Bifidobacterium та зменшення вмісту Candida spp. Екстракт гранату специфічно стимулює ріст Bifidobacterium. Існують значні відмінності у продукуванні цитокінів в супернатантах культури фрагментів тканин і в сироватці крові мишей при оральному введенні різних рослинних екстрактів. Екстракт кропу зумовлює системне і локальне збільшення ФНП-a та IЛ-2, а екстракти кропиви та Sideritis викликають таке збільшення лише на ділянках слизової оболонки. Екстракти гранату, хурми та капусти кале системно і локально стимулюють продукування ІЛ-2, ІЛ-10, ІЛ-12, ІФН-γ та ІЛ-17, але не ФНП-α.Висновки. Найбільш виразними корисними властивостями щодо впливу на стан кишкової мікробіоти та продукування цитокінів слизовими оболонками кишечнику володіють екстракт хурми, а також екстракти гранату та капусти кале. Extracts of Pomegranate, Persimmon, Nettle, Dill, Kale and Sideritis Specifically Modulate Gut Microbiota and Local Cytokines Production: in Vivo Study
The aim of the work was to use the in vivo model to reveal main changes in gut microbiota of immune competent mice in dynamic and find out the specificity of immunomodulation action (activity) of traditional foods ingredients - edible plants extracts on local mucosal cytokines to its oral administration.Materials and methods. In this study, seven groups of immunocompetent BALB/c mice were formed. All experimental mice had been fed orally by plants' extracts (15mg/200 µl/mouse) for the 14th days. The extracts of edible plants - ingredients of traditional food such as kale leaves, persimmon, pomegranate, dill, Sideritis scardica, and nettle were obtained as described and orally administrated to experimental animals. For microbiological analysis of gut microbiota changes, the colon content has been investigated, the key microbial representatives were isolated by plating of its serial dilution on selected chromogenic medium, identified serologically and biochemically. The production of cytokines in different gut compartments and gut associated lymphoid tissues (GALT) were detected by Enzyme-linked immunosorbent assay (ELISA).Results. In experiments in mice, the ability of Kale, Dill and Sideritis extracts, when administered orally, selectively inhibit the content of E. coli, K. pneumoniae, E. faecalis, L. acidophilus in the colon of mice had been demonstrated, and increasing of B. bifidum had been observed. Self-Assembly of Molecular Brushes with Polyimide Backbone and Amphiphilic Block Copolymer Side Chains in Selective Solvents
Three-component molecular brushes with a polyimide backbone and amphiphilic block copolymer side chains with different contents of the “inner” hydrophilic (poly(methacrylic acid)) and “outer” hydrophobic (poly(methyl methacrylate)) blocks were synthesized and characterized by molecular hydrodynamics and optics methods in solutions of chloroform, dimethylformamide, tetrahydrofuran and ethanol. The peculiarity of the studied polymers is the amphiphilic structure of the grafted chains. The molar masses of the molecular brushes were determined by static and dynamic light scattering in chloroform in which polymers form molecularly disperse solutions. Spontaneous self-assembly of macromolecules was detected in dimethylformamide, tetrahydrofuran and ethanol. The aggregates size depended on the thermodynamic quality of the solvent as well as on the macromolecular architectural parameters. In dimethylformamide and tetrahydrofuran, the distribution of hydrodynamic radii of aggregates was bimodal, while in ethanol, it was unimodal. Moreover, in ethanol, an increase in the poly(methyl methacrylate) content caused a decrease in the hydrodynamic radius of aggregates. A significant difference in the nature of the blocks included in the brushes determines the selectivity of the used solvents, since their thermodynamic quality with respect to the blocks is different. The macromolecules of the studied graft copolymers tend to self-organization in selective solvents with formation of a core–shell structure with an insoluble solvophobic core surrounded by the solvophilic shell of side chains

Метою роботи було визначення вмісту біологічно активних речовин, а саме поліфенолів та антоціанів,в екстрактах плодів аличі, чорниці, йошти, черешні, сливи, червоної та чорної смородини, та дослідження впливу екстрактів плодів цих рослин на ріст та біоплівкоутворенняклінічних ізолятів "збудників ESKAPE"in vitro.Матеріали і методи. Антибіотикорезистентність штамів клінічного походження, а саме Enterococcus faecalis, Staphylococcus aureus, Klebsiella pneumoniae, Acinetobacter baumannii, Pseudomonas aeruginosa, Enterobacter cloacae визначали методом дифузії Кірбі-Бауера. Вміст біологічно активних речовин визначали методом тонкошарової хроматографії. Вплив ягідних екстрактів на вищезгадані ізоляти вивчали методом їх сумісного культивування. Здатність клінічних ізолятів до утворення біоплівки вивчали за допомогою спектрофотометричного методу з використанням кристалічного фіолетового, в якості барвника.Результати. Проаналізувавши результати чутливості до антибіотиків клінічних ізолятів, було встановлено, що вони стійкі до всіх антимікробних препаратів. Аналіз вмісту біологічно активних речовин екстрактів ягід показав, що вони містять велику кількість антоціанів та поліфенолів, та володіють антибактеріальними властивостями. Було встановлено, що ізоляти клінічного походження здатні до біоплівкоутворення, а відібрані нами екстракти ягід володіють здатністю інгібувати сформовані біоплівки ізолятами клінічного походження, а саме: Klebsiella pneumoniae та Pseudomonas aeruginosa.Висновки. Досліджуючи антибактеріальні властивості антоціанів та поліфенолів, екстрагованих з ягід, можна зробити висновок, що вони здатні гальмувати ріст не лише виділених нами планктонних форм штамів, але й пригнічують утворені ними біоплівк
Molecular Brushes with a Polyimide Backbone and Poly(ε-Caprolactone) Side Chains by the Combination of ATRP, ROP, and CuAAC
An approach to the synthesis of the novel molecular brushes with a polyimide (PI) backbone and poly(ε-caprolactone) (PCL) side chains was developed. To obtain such copolymers, a combination of various synthesis methods was used, including polycondensation, atom transfer radical polymerization (ATRP), ring opening polymerization (ROP), and Cu (I)-catalyzed azide-alkyne Huisgen cycloaddition (CuAAC). ATRP of 2-hydroxyethyl methacrylate (HEMA) on PI macroinitiator followed by ROP of ε-caprolactone (CL) provided a “brush on brush” structure PI-g-(PHEMA-g-PCL). For the synthesis of PI-g-PCL two synthetic routes combining ROP and CuAAC were compared: (1) polymer-analogous transformations of a multicenter PI macroinitiator with an initiating hydroxyl group separated from the main chain by a triazole ring followed by ROP of CL, or (2) a separate synthesis of macromonomers with the desirable functional groups (polyimide with azide groups and PCL with terminal alkyne groups), followed by a click reaction. Results showed that the first approach allows to obtain graft copolymers with a PI backbone and relatively short PCL side chains. While the implementation of the second approach leads to a more significant increase in the molecular weight, but unreacted linear PCL remains in the system. Obtained macroinitiators and copolymers were characterized using 1H NMR and IR spectroscopy, their molecular weight characteristics were determined by SEC with triple detection. TGA and DSC were used to determine their thermal properties. X-ray scattering data showed that the introduction of a polyimide block into the polycaprolactone matrix did not change the degree of crystallinity of PCL
Novel regular polyimide-graft-(polymethacrylic acid) brushes: Synthesis and possible applications as nanocontainers of cyanoporphyrazine agents for photodynamic therapy
International audienceAn approach to the synthesis of new regular graft-copolymers polyimide (PI)-graft-polymethacrylic acid is elaborated, including (1) synthesis of multicenter PI macroinitiators, (2) controlled ATRP of tert-butylmethacrylate on the prepared macroinitiators, and (3) protonolysis of tert-butyl ester groups of side chains of the resulting PI-graft-poly(tert-butylmethacrylate). Experimental conditions for attaining complete conversions of the first and the third stages of the process are determined by means of 1H NMR and FTIR-spectroscopy. Polymer products of the first and the second stages of the process, as well as poly(tert-butylmethacrylate) side chains cleaved from the PI-graft-poly(tert-butylmethacrylate) copolymers by complete decomposition of the PI backbone under alkaline hydrolysis conditions, are characterized by GPC. The kinetics of poly(tert-butylmethacrylate) chain growth on a PI macroinitiator under ATRP conditions are studied. The results obtained provide evidence for the controlled character of the ATRP process and the regular structure of the synthesized graft-copolymers.
